“You can start, even in old age, the function of stem cells,” Rando said. “We end up concluding that, if these studies are generalizable, then we see that exercising old mice would also be applicable to humans.”
Exercise reprograms the inflammatory landscape of multiple stem cell compartments during mammalian aging
Exercise has the ability to rejuvenate stem cells and improve tissue regeneration in aging animals. However, the cellular and molecular changes elicited by exercise have not been systematically studied across a broad range of cell types in stem cell compartments. We subjected young and old mice to aerobic exercise and generated a single-cell transcriptomic atlas of muscle, neural, and hematopoietic stem cells with their niche cells and progeny, complemented by whole transcriptome analysis of single myofibers. We found that exercise ameliorated the upregulation of a number of inflammatory pathways associated with old age and restored aspects of intercellular communication mediated by immune cells within these stem cell compartments. Exercise has a profound impact on the composition and transcriptomic landscape of circulating and tissue-resident immune cells. Our study provides a comprehensive view of the coordinated responses of multiple aged stem cells and niche cells to exercise at the transcriptomic level.

Why Former Google CEO Eric Schmidt Is Betting Big On The $1T US Bioeconomy
After being at the helm for over a decade of internet juggernaut Google, former CEO Eric Schmidt has switched gears and one of his latest activities is being an advocate for the fast-growing US bioeconomy, which is valued at over $1T.
In biotech, companies like Genentech and BioNTech have demonstrated how years of lab research can turn into solutions that scale to serve billions.
“I think it’s time. It’s time to take the work of the last 10 or 20 years and make it scale to a global phenomenon that is 4-30 trillion dollars. You all will do that. That’s why I’m here.” Schmidt explained to a packed house.
Interestingly, Schmidt made it clear that to stop local companies from outsourcing their operations, the US needs to build a national, shared infrastructure that boosts the domestic bioeconomy. Put another way, the US needs to build an official governmental home for bioeconomy interests.
From a regulatory standpoint, the Bioeconomy Executive Order put into place by the Biden administration last September will systematically shift how the US coordinates and strategically advances its bioeconomy goals toward multiple objectives, including carbon management, economic development, biosecurity, biosafety, and the workforce of the future.
“One can hope that these actions will include a deliberate focus on moving engineering biology discoveries from lab to market, helping new technology developers through new loan programs for biomanufacturing infrastructure, as well as through newly streamlined, clear regulatory processes,” Schmidt remarked. /
Synergistic Effect of Metformin and Lansoprazole Against Gastric Cancer through Growth Inhibition
Cancer has been linked to metabolic disorders and diverse gene mutations. Metformin, which is widely used to treat type 2 diabetes, inhibits the growth of cancer cells in animal models. Here we investigated the effects of metformin on human gastric cancer cell lines. We also investigated the synergistic anticancer effect of metformin and proton pump inhibitors. Lansoprazole, a proton pump inhibitor, is effective for treating gastroesophageal reflux disease. Our results revealed that metformin and lansoprazole can significantly inhibit cancer cell growth in a dose-dependent manner by suppressing cell cycle progression and inducing apoptosis. Low concentrations of metformin and lansoprazole have a synergistic effect on AGS cell growth inhibition. In summary, our findings suggest a new and safe treatment protocol for treating stomach cancers.
Cell morphology
To observe the real-time images of the cells after drug treatment, we captured time-lapse images using HoloMonitor M4 (Phase Holographic Imaging AB, Lund, Sweden). The cells were seeded in a 6-well plate at 5000 cells/well and incubated overnight. On the next day, the plate with hololid were placed on the HoloMonitor M4 after the drugs were added. The cell images were recorded every 20 min for 72 h. Finally, the holographic images were analyzed using the HoloMonitor M4 application program.
