For the cell migration trajectory assay, A549 and PC-9 cells were seeded (2  105 cells/well) in a 6-well plate. After 12 h, the cells were monitored using HoloMonitor M4 (Phase Holographic Imaging PHI AB, Skiffervägen, Lund, Sweden) and photographs were taken every 5 minutes for 20 minutes. 3.5. Fazna holografska mikroskopija na mioblastima Nakon 24 sata pripremila sam HoloLid™ kako bih ga postavila na pločicu sa nasađenim mioblastima uz pomoć protokola proizvođača (Phase Holographic Imaging PHI, Švedska). Nakon što sam pažljivo provjerila da se u potpunosti osušio nakon sterilizacije, HoloLid™ sam postavila na pločicu s mioblastima, a pločicu smjestila na HoloMonitor® M4. Uređaj HoloMonitor® M4 koristila sam za neinvazivno praćenje stanica u stvarnom vremenu (Slika 7). Ovaj uređaj omogućuje holografsku mikroskopiju, odnosno 2D i 3D analizu mioblasta, uključujući promjene u obliku i površini stanica, njihovu diobu, migraciju i formiranje miotuba. HoloMonitor® M4 generirao je slike svakih 30 minuta na 10 različitih pozicija unutar svake jažice tijekom 72 h unutar CO2 inkubatora (5 % CO2, 37 °C). Nakon završetka snimanja, pločicu sa stanicama uklonila sam s uređaja i ostavila u CO2 inkubatoru (5 % CO2, 37 °C).

Unraveling Ammonia-Induced Brain Endothelial Senescence: Role of miRNA-183-5p 4 August 2025

First, we observed reduced by ∼20% RBE4 cell proliferation and captured by the live cell HoloMonitor recording of cell number. Results corroborate the observation from our laboratory, which demonstrated ammonia-induced RBE4 cell reduction with an MTT test assessing cell viability (Skowrońska et al., 2012). After seeding the cells were left to adhere for 4–6 hours, then washed with PBS. New DMEM was added, and the plates were inserted to the Holomonitor M4, which uses quantitative phase imaging to automatically detect and measure live cells without labelling.

The speed of individual cells between each consecutive captured image frame (distance travelled/time between frames) was calculated by the built-in software of the HoloMonitor, as well as the average speed of all cells of each type between consecutive frames.

2.9. Time-Lapse Cell Tracking Analysis

For live cell migration tracking, MEF cells were seeded at a density of 9.0 × 103 cells per 100 μL in DMEM lacking fetal bovine serum and supplemented with 2% Matrigel, using μ-Slide I chambers (Ibidi, #80106). After a 3 h pre-incubation period, 900 μL of DMEM containing 10% FBS was gently added to each well, followed by overnight incubation. Cells were subsequently treated with 100 μM of compounds 14 and immediately transferred to a HoloMonitor M4 imaging system (phase-holographic cytometry) maintained at 37 °C. Time-lapse images were acquired at 15 min intervals over a 12 h period, and cell motility was quantified using HoloStudio M4 analysis software (v2.6.4; Hstudio M4, Sun Valley, CA, USA).

4.3. Quantitative Analysis of Cell Morphology and Motility

The In-depth Analysis: “Cell Morphology” module of the Holomonitor App Suite (PHI, Sweden) was used to track the movement of single cells. The following cell identification settings were used: automatic minimum error method, 134 adjustments, and minimum object size 20. Measurements of optical thickness, optical volume, roughness, shape convexity, irregularity, and motility were performed at specific time points. Using the “Kinetic Assays” module: “Kinetic Motility Assays”, In-depth Analysis: “Wound Healing Assay” was studied.

To investigate cellular migration traits in detail, we employed the HoloMonitor® M4, an automated live-cell imaging cytometer that performs time-lapse analysis. This system allows for the imaging and quantification of unstained, living cells directly within their culture vessels inside the incubator, enabling real-time monitoring of various morphological and migratory parameters.

Kvantitativ fasmikroskopi

För att mäta cellernas torrvikt använde vi Holomonitor M4 kvantitativt fasmikroskop (PHI). QPM bygger på principen att brytningsindexet för ett material omvänt korrelerar med hastigheten hos ljuset som passerar genom det. När Holomonitor är i drift riktar den en laserstråle genom både cellen och det omgivande odlingsmediet. När ljus passerar genom cellen, som har ett högre brytningsindex än mediet, saktar det ner, vilket resulterar i en detekterbar fasförskjutning. Denna fasförskjutning fångas av kameran och bearbetas för att sammanfatta den totala fasförändringen som orsakas av hela cellen (optisk volym). Den totala fasförändringen av enskilda celler analyserades med hjälp av den dedikerade programvaran Holomonitor M4.

3.2.1 CELL VIABILITY ASSAYS AND HOLOMONITOR LIVE CELL MONITORING
To enable continuous monitoring of treatment response, a HoloMonitor live cell imaging system was used. This system employs laser-based quantitative phase imaging to assess cell number, confluence, and volume, without the need for chemical reagents (Figure 13). By eliminating reagent use, potential biases such as toxicity or uneven loading are avoided. Additionally, the system allows for real-time monitoring of cell lines prior to extraction and subsequent RNA and protein analyses. However, these experiments are more time consuming and require specialized holographic lids, as well as careful data interpretation and analysis to ensure reliable results. At the end of most HoloMonitor-based cell viability assays, alamarBlue was added to provide complementary data on metabolic viability.

2.6. Time-laps videos

Time lapse images were captured with a HoloMonitor® M4 digital holographic microscope with a motorized stage (M4) (PHI AB, Lund, Sweden) placed in a regular 37 ​°C CO2 incubator with 5 ​% CO2 in air. For time lapse-imaging of cells in the M4 microscope, cells were seeded in 6-well plates. The cells were allowed to attach for 24 ​h prior to start of the experiment. The standard lid of the Petri dish was replaced with a HoloLid® 71120 for 6-well plates (PHI AB). Cells were treated with 100 or 1000 ​μM lidocaine or ropivacaine. Control cells received PBS. The 6-well plate was then placed on the stage of the M4. The M4 is a quantitative imaging system based on digital holographic microscopy. When imaging with the M4 microscope, the software HoloMonitor® AppSuite was used. Images were acquired at 6–8 positions per well, with 5 ​min intervals for 72 ​h.

Digital holographic microscopy (DHM) HaCaT cells were seeded into one 6-well plate from SARSTEDT with a cell concentration of 16’000 cells and left to adhere overnight before being replaced with EPILIFE medium and 1 and 10 mM NIA to get a final volume of 5 mL. Antibiotic with a concentration of 0.125 µg/mL amphotericin B from Thermo Fisher, Life Technologies, Gibco (Paisley, UK) was also added, the lid was replaced with HoloLids™ (Phase Holographic Imaging AB), and then placed on the DHM, HoloMonitor™ M4 (Phase Holographic Imaging AB, PHIAB, Lund, Sweden). The instrument was then calibrated for 10 minutes, incubated at 37°C with 5% CO2. Using the AppSuite™ software (Phase Holographic Imaging AB) of the instrument, 10 random positions were generated and left running for 72 hours in the same incubator in which it was calibrated, with images taken every two hours. Once the photos were taken, the software counted the number of cells and provided the cell count in x/cm2 and the confluency over time.

2.7 Phase holographic microscopy

To study cell behavior on different types of nanosubstrates, cells were imaged using a phase holographic microscope (HoloMonitor, Phase Holographic Imaging PHI AB). The microscope and associated software (HoloMonitor App Suite) give 3D information about the cells without any perturbations since the measurements are performed in the incubator on unlabeled cells.

3. Results

3.1. MHV-JHM Replication in bEnd.3 Cell Line

To identify a viral dose suitable for establishing long-term, non-cytolytic MHV-JHM infection in bEnd.3 cells, we monitored the proliferation of cultures infected with serial 10-fold dilutions of virus stock (100 to 10−5) over 72 h using HoloMonitor® PHI M4 (Phase Holographic Imaging, Lund, Sweden) live-cell imaging. Uninfected controls were included for comparison. Cell counts were recorded at 0 h and 72 h p.i., and statistical analyses were performed to assess both within-group changes and differences between conditions

Det kan konstateras att de forskande HoloMonitoranvändarna världen över inte har legat på latsidan inför/under sommaren. Ovanstående är ett axplock som jag snabbt hittade.Den som vill ha in fler nya studier till den här listan kan mejla över dem så lägger jag till dem.

  3. 20st ja... Det är verkligen svårt att förstå hur alla dessa rapporter inte kan ge större effekt på säljet. Är det bara rapporter utan praktiskt värde? Men jag har å andra sidan ingen annan erfarenhet än PHI på området heller. Heja säljorganisationen och kommande cellsync! Hoppas det släpper nu
