Lösa trådar

 I min nätsökning på PHI-relaterat kom jag till en sida som tar upp patent. Där fanns en del intressanta saker som är nära i tiden. Hur man ska värdera informationen vet jag inte. Kanske nån brajt phi,are kan sprida lite ljus här? Samtliga patent har en koppling till HoloMonitor och PHI.

1. Boston Children Hospital har ett pending patent sen Juli, på ett nytt sätt att behandla barn med bukspottscancer/pancreas (som jag förstår det). Länk

HoloMonitorkoppling : Therapeutic effect of ICAM-1 Ab

The therapeutic effect of ICAM1 antibody on human pancreatic cancer cell lines in vitro was assessed using quantitative phase contrast imaging. IgG was used as a control. Cells were seeded at a density of 5× 104 cells/well in 6-well plates. After 24 hours of recovery, the entire medium was replaced with that containing ICAM1 antibody or IgG at a dosage of 2 μg/mL. Cells were incubated in ICAM1 or IgG-containing medium at 37°C for 24 hours. Plates were then placed under a quantitative phase contrast imaging microscope (Holomonitor M4, Phase Holographic Imaging Phi AB, Lund, Sweden) in an incubator and imaged at 5 min intervals for another 24 hours. Cell motility, morphology, and proliferation were then analyzed using Hstutio4.

HoloMonitorkoppling : The culture solutions of M0 macrophages, M1 macrophages, and M2 macrophages were placed on the sample stage of a holographic microscope (HoloMonitor M4 manufactured by PHI AB) and cultured for 36 hours in a CO2 incubator while time-lapse photography was performed at 30-minute intervals. The number of samples photographed was 7991 for M0 macrophages, 8129 for M1 macrophages, and 7103 for M2 macrophages.

From the obtained images, software (HoloMonitor M4 App Suite manufactured by PHI AB) was used to obtain the above-mentioned "Area," "Irregularity," "Optical volume," and "Optical thickness" as parameters indicating cell morphology, along with the time elapsed since cell division (Time), and the above-mentioned "Migration distance," "Motility distance," and "Motility speed" as parameters indicating changes in cell position over time.

The culture plate containing undifferentiated cells and the culture plate containing differentiated cells were placed on the sample stage of a holographic microscope (HoloMonitor M4 manufactured by PHI AB), and while culturing for 36 hours in a CO2 incubator, time-lapse photography was performed at 30-minute intervals. The number of samples photographed was 5,356 for undifferentiated cells and 10,328 for differentiated cells.

3. Gabriel Nussbaum (privatperson/forskare?) fick i juli patent på nåt slags detekteringsinstrument för bakterier/cancer. Länk Tillägg Gabriel är en amerikansk professor som flyttat till Israel där han bedriver cancerforskning. Mer info här.

HoloMonitorkoppling : Live cell imaging: A HoloMonitor M4 digital holographic cytometer (DHC) from Phase Holographic Imaging (PHI, Lund, Sweden) was placed in a Don Whitley H35 Hypoxystation (don whitley Scientific) maintained at 94%N2/5%CO2/1%O2 and 37°C. Timelapse phase imaging, as well as image processing, segmentation, and analyses, were performed using the Hstudio software package (PHI). Cells were plated in 96 well plates and each well was imaged in 3-5 areas every 60-90 minutes over 48-72 hours. The Cell Count module in HStudio was used to segment the phase images per well, followed by the object identification and cell counting modules in HStudio. Multiple images of the same gap and multiple wells were analyzed to assess reproducibility.

HoloMonitorkoppling : Cell migration was also examined using a digital holographic microscope (Figure 11). A HoloMonitor (R) M4 microscope (Phase Holographic ImagingAB ) was used to image and track the movement of XP-A-iMCs and HC-iMCs after high-dose (150 J/ m2 ) UV-B irradiation. XP-A-iMCs and HC-iMCs ( 1x104 cells) were seeded in a 6-well plate, and migration was tracked for 72 hours after high-dose (150 J/m2) UV-B irradiation. Figure 12 shows the distance traveled by the cells, and Figure 13 shows the speed of cell migration. It was confirmed that migration ability was reduced in XP-A-iMCs with reduced HDAC4 after UV-B irradiation. This suggests that the phenotype of XP-A-iMCs may cause pigmentation and depigmentation, which are characteristic skin symptoms in XP-A patients. Furthermore, since Example 2 showed that the survival rate of XP-A-iMC decreased due to light exposure, it may be possible to obtain a substance that improves cytotoxicity in melanocytes by adding a candidate substance to a melanocyte culture system and evaluating the survival rate and migration ability of melanocytes.
